The Bioreactor Bag Manufacturing and Reuse Kit (RBBK2) is a solution inspired by bag technology as used in Gen-1 and Gen-2 bioreactors and a late-stage evolution of the original RBBK. The development of bioprocesses in space missions faces several technical and logistical challenges. Gravity on Earth allows phase separation at levels, facilitating an orderly movement of fluids. In microgravity, the dominant force is viscosity, causing liquids and gases to separate into bubbles. The liquid of interest is not always positioned at the inlet/outlet ports, which makes its operation extremely diicult, especially when using rigid vessels. The proposed new RBBK2 design introduces significant improvements in terms of reusability, on-site manufacturing and compatibility with existing infrastructure. Its modular design allows it to be adapted to dierent uses beyond bioreactors, maximizing eiciency and reducing the need for disposable consumables. It consists of two main subassemblies and a 3D printed inner scraper: Subset bag (10-SubSet-bag.stp) : Made of gas-permeable plastic, it allows for eicient exchange of 𝑂, 𝐶𝑂and ethylene. Luer Lock Valve Subset (20-SubSet_luer-lock.stp) : Facilitates the injection and withdrawal of fluids through a LUER LOOCK port Scraper (30-scraper.stp) : Allows you to clean the inside of the bags without having to open them. Each component has been designed to use appropriate materials for its function and to be manufactured with the 3D printer available on the ISS. The simple and lightweight structure of RBBK2 (less than 13 g), together with its reusability and recyclability, make it a very viable option for building bioreactors capable of handling liquid content in microgravity environments during space missions.
